该班机原定计划于北京时间（UTC+08:00）6:30抵达北京首都国际机场，但起飞后不足一小时便在马来西亚与越南海域的交界处、土珠岛（又译土朱岛）以南约140海里及哥打巴鲁东北东约90海里处与马来西亚梳邦空管中心失去联系。

随后，根据数据分析，推定的飞机失踪位置被多次修改，最终认为航机进入并坠毁于南印度洋。2015年1月29日，即客机失踪10个月后，马来西亚民航局代表马来西亚政府正式宣布航班失事，机上239人全数罹难。

2015年7月29日，法属留尼旺岛寻获一块疑似大型飞机襟副翼残骸，上面有一组波音专属的维修编号，经初步丈量判断属于777型客机零件，被送往法国土鲁斯国防部装备总局航空技术中心(DGA Techniques aéronautiques)实验室分析，于2015年8月6日被马来西亚首相纳吉间接证实属于MH370。

9月3日，法国巴黎检察院宣布结果，确定该样本是编号9M-MRO的波音777机翼部件。由澳大利亚主导的水下搜救工作现仍在继续。直至2016年，该空难为商业航班空难事故史上，唯一未能确定具体失事地点的事故。
